高血脂是导致心脑血管疾病、脂肪肝、胰岛素抵抗等各类代谢性疾病的重要危险因素。营养过剩及不良生活习惯是诱发高血脂的重要原因,而不同人群对这些诱因的敏感程度有很大差别,这主要是由遗传因素决定。据估算,50%以上的血脂异常可归因于遗传因素。我们实验室主要感兴趣的是揭示人群中血脂异常发生发展易感性背后的遗传与分子机制,结合生物化学、细胞生物学和实验动物模型等手段阐明其发病机制与作用原理,为代谢性疾病的诊治提供理论基础。
我们前期的工作揭示了肝脏分泌性蛋白PCSK9,ANGPTL3,ANGPTL8等调节脂质代谢的功能与机制,尤其是ANGPTL3-ANGPTL8是首次发现的调节组织特异性脂肪摄取的关键蛋白质复合体,对研究组织间的信号通讯、脂质代谢的生理调控及脂肪的异位堆积等生理病理过程具有重要的意义。针对PCSK9的抑制剂已进入临床,成为新一代降胆固醇药物,针对ANGPTL3的抑制剂已进入三期临床试验,取得良好降血脂效果。我们近期的工作发现一个非编码区致病单碱基突变,并揭示其通过上调GPCR孤儿受体GPR146的转录表达,导致高胆固醇血症的作用机制,这也是首次发现的直接调节血液胆固醇水平的GPCR,为后继药物研发奠定关键基础,相关工作发表在Cell Metab,Cell Res,PNAS,J Cell Bio等期刊,他引2000余次。
